Natural drugs in cancer treatment and their cytotoxicity

The thesis deals with the natural substances in context with the cancer disease. The natural substances have a positive effect on the human organism and they are able to influence the viability and the growth of the cancer cells. The main mechanical device is to influence the mechanisms needed to start the apoptosis of the cancer cells and stopping further proliferation. The cancer cell lines utilization in the cancer disease is discussed in the thesis too. The thesis states common methods of determining the natural substances cytotoxicity. For the experimental part of the thesis it was chosen the MTT test method and the xCELLigence system for monitoring in real time. The mechanical device of the tested substance capsaicin in application on the prostate cell lines, tumorous PC3 and nontumorous PNT1A influence will be observed within the experimental part of the thesis.
Effect of cancer therapy on fertilization parameters in testicular germ cell tumour patients.

Testicular germ cell tumours (TGCTs) are a rare type of cancer. It is the most common type of cancer in the age group of 15-35 years. The consequences that TGCT and the treatment of this disease impose to these young men in terms of sperm parameters and fertility is yet unclear. This study involves 84 semen samples from 46 patients sorted in three time groups - before treatment, 3 months after the treatment and 6 months after the treatment. Several sperm parameters were studied and compared within the three time points. Adding on to this, the study takes two patients with complete set of semen samples and studies them individually. The results mostly show great differences in patients and this study underlines the individual characteristics that need to be taken into consideration when it comes to patients' fertility journeys after battling TGCT. Sperm viability, concentration, apoptosis, DNA damage, acrosomal integrity and leukocyte concentration nor histone modification evaluation showed any significant changes between said groups. DNA damage was correlated to the level of acrosomal permeability. IgA antibody levels rose significantly in samples after orchiectomy. IgG antibody levels did not show the same trend. Theragnostics - new methods in nuclear medicine

The main topic of the bachelor thesis is theragnostics - new methods in nuclear medicine. Theragnostics is a personalized treatment strategy that combines diagnostics with therapy and finds application primarily in the treatment of cancer. Theragnostics as a medical field is still evolving. The aim of this work was to map the practical use of current indications of theragnostic methods in nuclear medicine. The theoretical part of the thesis defines the concept of theragnostics and theragnostic methods containing nanoparticles and radionuclides. There is described the use of radionuclide theragnotics in clinical practice, especially in the treatment of thyroid gland, neuroendocrine tumors and prostate cancer. In this part of the thesis there are also clinical studies for theragnotic practice. Furthermore in the theoretical part of the thesis is maped the practical use of current indications of theragnotic methods, as well as possible future perspectives of theragnotic treatment. The theoretical part is processed from professional publications and articles. The practical part of the work focuses on the awareness of radiology assistants about this new treatment method. The respondents were radiology assistants across hospitals. There were 32 respondents involved to complete the research questions for the questionnaire survey. The evaluation from the collected data of the questionnaire survey is in the form of the percentage success of radiological assistants. The overall concept of this bachelor's thesis can serve as a study material for university students in the field of radiology assistant.
Molecular mechanisms of apoptosis induction in cancer cells.

Cancer diseases are now the third leading cause of death (20% of all deaths). It is therefore important to find new ways of getting tumor cells effectively and specifically disposed of and a promising path is targeted therapy. One of the most frequently deregulated protooncogenes in cancer is C-MYC, which makes it suitable as an effective target for treatment. However, the development of such targeted active ingredients is very expensive, so in this thesis we investigate natural substances that have been used for the treatment of cancer in ancient China. We examined the substances shikonin, cnicin and artemisinin. The results show that shikonin induces apoptosis of tumor cells by reducing the expression of C-MYC and activating tumor suppressor kinase MST1. Cnicin reduces the expression of C-MYC as well, but activates MST1 only weakly. Artemisinin, on the other hand, increases expression of C-MYC and doesn't activate MST1, thus operates on inducing apoptosis of tumor cells by a completely different mechanism. (In Czech)
Tumor suppressor NDRG1 and its regulation by iron chelators

Iron is an essential trace element required for many processes within a cell, including DNA synthesis and cell cycle progression. Moreover, it is critical for cellular respiration in mitochondria. Due to their proliferative nature, cancer cells are dependent on iron, and depleting this element via iron chelators results in the inhibition of ribonucleotide reductase, leading to cell cycle arrest and apoptosis of cancer cells. Recently, an alternative mechanism for the effect of iron chelators have been proposed, including induction of N-myc downstream regulated gene 1 (NDRG1) expression and its inhibitory effect on c-MET, EGFR, and NF-κB pathways, which can act as oncogenes in a certain context. NDRG1 is a tumour suppressor gene, which is downregulated in many cancers and its downregulation correlates with cancer progression, poor differentiation, and higher metastatic potential. It has been shown that NDRG1 expression can be regulated by intracellular iron - a decrease in intracellular iron leads to upregulation of NDRG1 at mRNA and protein level via the HIF-1-dependent mechanism by inhibiting prolyl hydroxylases. Male infertility in context of testicular cancer

This bachelor thesis focuses on male infertility in a connection to testicular cancer. Testicular cancer is the most common malignancy among young men in a reproductive age and the worldwide incidence of testicular cancer is on the rise. A lot of attention is also given to an increasing rate of infertility in a context of testicular cancer. For these reasons, the aim of this thesis is to clarify several non-physiological changes, such as hormone levels, spermatogenesis and sperm parameters, which take place in a male body and influence the chance to become a biological father. These pathological changes can be due to the disease itself but also due to the treatment. A cryopreservation of semen is also going to be discussed as the way to preserve male fertility and as an option for couples with the infertility problems due to male factor which rely on help of the assisted reprodiction.
